Drug intelligence / Profile preview
POU2AF1 (BOB-1) inhibitors are a class of therapeutic agents designed to target and inhibit POU class 2 homeobox-associating factor 1 (POU2AF1), also known as B-cell-specific transcription coactivator OCA-B, OBF-1, or BOB-1. POU2AF1 is a transcriptional coactivator that plays a critical role in B-cell development, germinal center formation, and immunoglobulin production, and is highly expressed in multiple myeloma cells and other B-cell malignancies. In T cells, POU2AF1 interacts with the transcription factor Oct1 to regulate the expression of immunomodulatory cytokines, making it a key driver of autoimmune diseases such as type 1 diabetes and Sjögren's syndrome. Therapeutic strategies targeting POU2AF1 include small molecule inhibitors (such as DB1033) that disrupt the Oct1:OCA-B:DNA complex, antisense oligonucleotides (such as gapmers) that downregulate POU2AF1 expression, and membrane-penetrating peptide inhibitors. These agents are being investigated for the treatment of multiple myeloma, B-cell lymphomas, and autoimmune disorders.
