This is a combination immunotherapy consisting of an autologous dendritic cell (DC) vaccine pulsed with mRNA encoding the human cytomegalovirus (CMV) matrix protein pp65 fused to lysosome-associated membrane protein (shLAMP), administered together with granulocyte-macrophage colony-stimulating factor (GM-CSF). The DCs are engineered ex vivo to express the CMV pp65 antigen targeted to lysosomes via shLAMP, enhancing antigen presentation on MHC-II and stimulating T-cell mediated immune responses against tumor cells expressing CMV antigens. GM-CSF acts as an adjuvant, promoting differentiation and activation of DCs and other immune cells at the vaccination site. This combination has been investigated primarily for glioblastoma multiforme (GBM), aiming to induce robust anti-tumor immunity in patients[2][3][6][8].
