Overview

This preclinical-stage small molecule program, developed by the Global Alliance for TB Drug Development (TB Alliance), targets the enzyme phosphopantetheinyl transferase (PptT) for the treatment of tuberculosis. PptT is an essential enzyme in *Mycobacterium tuberculosis* that catalyzes the transfer of a 4'-phosphopantetheine moiety from coenzyme A to various carrier proteins. This post-translational modification is crucial for the activation of enzymes involved in the biosynthesis of mycolic acids and other complex lipids essential for the bacterial cell wall and virulence. By inhibiting PptT, the program aims to disrupt bacterial cell wall integrity and lipid metabolism, providing a novel mechanism of action against both drug-sensitive and drug-resistant tuberculosis strains. The program is currently in the discovery and lead identification phase of development.
