Combination of **PQR309**, an oral, balanced pan-PI3K/mTOR inhibitor, and **eribulin (Halaven)**, a microtubule dynamics inhibitor, studied as a therapeutic regimen for patients with locally advanced or metastatic HER2-negative and triple-negative breast cancer. PQR309 inhibits both PI3K and mTOR signaling pathways, which are frequently activated in cancers, thereby suppressing downstream signaling critical for tumor progression. Eribulin inhibits the growth phase of microtubule dynamics, leading to cell cycle arrest and apoptosis in cancer cells. Preclinical and early clinical results suggest potential synergistic activity for this drug combination in aggressive breast cancer subtypes, notably triple-negative breast cancer.
