Drug intelligence / Profile preview
PR-3 is an ophthalmic nanoemulsion formulation of cyclosporine developed by Puripharm for the treatment of dry eye disease and severe keratitis. Cyclosporine is a small molecule calcineurin inhibitor that works by binding to cyclophilin A, forming a complex that inhibits the phosphatase activity of calcineurin. This inhibition prevents the dephosphorylation and subsequent nuclear translocation of the nuclear factor of activated T-cells (NFAT), thereby reducing the production of pro-inflammatory cytokines such as interleukin-2 (IL-2). The nanoemulsion formulation is designed to enhance the solubility and ocular surface retention of the hydrophobic cyclosporine molecule, potentially improving therapeutic efficacy and patient comfort compared to traditional oil-based formulations.
