Overview

Pramlintide + sibutramine is an investigational combination therapy for obesity and weight management. Pramlintide is a synthetic analogue of amylin, a hormone co-secreted with insulin by pancreatic beta cells, which acts to increase satiety and reduce food intake. Sibutramine is a centrally acting serotonin-norepinephrine reuptake inhibitor that suppresses appetite by increasing levels of these neurotransmitters in the brain. The combination targets distinct regulatory pathways involved in energy balance and appetite control, aiming for additive or synergistic effects on weight loss. Clinical studies have shown that this combination leads to significantly greater weight loss compared to either agent alone or placebo over 24 weeks in overweight or obese individuals[1][2][5][6]. However, use of sibutramine has been associated with increases in heart rate and blood pressure.
