The pre|CISION dual warhead compounds are a novel class of peptide drug conjugates (PDCs) developed by Avacta Therapeutics. This platform utilizes a proprietary peptide substrate that is specifically cleaved by Fibroblast Activation Protein alpha (FAP), a post-proline protease highly overexpressed on cancer-associated fibroblasts in more than 90% of solid tumors. Unlike traditional single-payload conjugates, these compounds are engineered to deliver two complementary cytotoxic or targeted agents simultaneously from a single molecule. Upon encountering FAP in the tumor microenvironment, the peptide linker is hydrolyzed, releasing both payloads directly at the tumor site. This dual-payload approach, exemplified by programs like AVA6207, aims to enhance therapeutic efficacy, overcome drug resistance, and minimize systemic toxicities such as neutropenia by ensuring the active warheads remain inert until they reach the tumor.
