Overview

CRx-119 is a novel orally available syncretic drug candidate developed for immunoinflammatory diseases. It is a fixed-dose combination of low-dose prednisolone (a glucocorticoid steroid) and amoxapine (a tricyclic antidepressant). The rationale behind this syncretic approach is to achieve synergistic therapeutic effects through multiple pathways, amplifying the anti-inflammatory and immunomodulatory actions of prednisolone while minimizing steroid-associated toxicity. Prednisolone acts as a glucocorticoid receptor agonist, inhibiting leukocyte infiltration, suppressing inflammatory mediators such as prostaglandins and leukotrienes, and modulating gene expression involved in immune responses. Amoxapine contributes by inhibiting the reuptake of norepinephrine and serotonin, which may further modulate inflammation. CRx-119 was primarily investigated for chronic adult periodontitis but was considered for broader immunoinflammatory conditions[1][2][3][4].
