prednisolone + dipyridamole

This is a fixed-dose oral combination drug consisting of the glucocorticoid prednisolone and the antiplatelet agent dipyridamole. Developed as Synavive (CRx-102), it was designed to enhance anti-inflammatory efficacy while minimizing glucocorticoid-associated side effects by using a very low dose of prednisolone synergistically amplified by dipyridamole. The mechanism involves both immunosuppressant and anti-inflammatory actions, with dipyridamole acting as a cyclic GMP phosphodiesterase inhibitor and nucleoside transport protein inhibitor, while prednisolone acts as a glucocorticoid receptor agonist. The combination was primarily investigated for inflammatory diseases such as rheumatoid arthritis, osteoarthritis, and other steroid-responsive conditions[3][6][7]. Clinical studies showed that this combination could reduce disease activity in rheumatoid arthritis with fewer side effects compared to higher doses of steroids alone[5]. Development has been discontinued.
