The combination of **prednisone**, **cytarabine**, and **vincristine** is a chemotherapy regimen developed and evaluated by **Shanghai Children's Medical Center** for the treatment of newly diagnosed multisystem Langerhans cell histiocytosis (MS-LCH) in children. This protocol was designed as a modification of the international LCH-II (Arm B) study, specifically investigating the efficacy of adding cytarabine to the standard induction backbone of prednisone and vincristine. Prednisone is a synthetic glucocorticoid that provides anti-inflammatory and immunosuppressive effects; cytarabine is an antimetabolite that inhibits DNA polymerase and interferes with DNA synthesis; and vincristine is a vinca alkaloid that inhibits microtubule formation, leading to mitotic arrest. The regimen aims to improve response rates and reduce disease reactivation, particularly in patients with or without risk organ involvement.
