prednisone + everolimus

Prednisone + everolimus is a combination therapy consisting of the mTOR inhibitor everolimus and the corticosteroid prednisone. This combination was primarily investigated in a Phase II clinical trial for patients with metastatic renal cell carcinoma (mRCC) who had progressed on prior VEGF-targeted therapies. The rationale for the combination was to utilize the anti-inflammatory properties of prednisone to mitigate common everolimus-induced adverse events, specifically noninfectious pneumonitis (NIP) and stomatitis. However, the trial was terminated early due to an unexpectedly high incidence of NIP, which occurred in 50% of the study population. Beyond oncology, the combination is also utilized in transplant medicine as part of immunosuppressive regimens for the prophylaxis of organ rejection.
