Prednisone is a synthetic glucocorticoid that exerts broad anti-inflammatory and immunosuppressive effects by activating the glucocorticoid receptor, leading to the modulation of gene expression for various inflammatory mediators. Iguratimod is a small-molecule disease-modifying antirheumatic drug (DMARD) that inhibits the NF-κB signaling pathway, thereby suppressing the production of pro-inflammatory cytokines such as IL-1, IL-6, and TNF, and inhibiting immunoglobulin production by B cells. This combination regimen is designed to induce and maintain remission in IgG4-RD, a systemic fibro-inflammatory condition characterized by tumefactive lesions and elevated serum IgG4 levels.
