Overview

This maintenance therapy regimen consists of the synthetic glucocorticoid prednisone and the recombinant cytokine interferon alfa-2b. It was evaluated by the PETHEMA Foundation (Grupo Español de Mieloma) in the GEM 2000 Phase 3 clinical trial as a post-autologous stem cell transplantation strategy for patients with symptomatic multiple myeloma. Prednisone acts as an anti-inflammatory and lymphocytotoxic agent by binding to glucocorticoid receptors, while interferon alfa-2b exerts antiproliferative, immunomodulatory, and anti-angiogenic effects by binding to the Type I interferon receptor (IFNAR). The regimen was designed to prolong remission and improve survival outcomes in patients who achieved a response following induction chemotherapy and high-dose melphalan conditioning.
