Pregabalin + amitriptyline is a fixed-dose combination (FDC) small molecule therapy developed for the management of neuropathic pain. It combines pregabalin, an anticonvulsant that binds to the alpha-2-delta (α2δ) subunit of voltage-gated calcium channels to reduce excitatory neurotransmitter release, with amitriptyline, a tricyclic antidepressant (TCA) that inhibits the reuptake of serotonin and norepinephrine. By targeting both the ascending pain transmission and descending inhibitory pathways, the combination aims to provide superior analgesic efficacy compared to monotherapy. While Arrier Biotech is listed as a distributor of this combination, it is widely available as a generic product in various markets, particularly in India, for conditions such as diabetic peripheral neuropathy and postherpetic neuralgia.
