Overview

Pregabalin + tramadol is a combination of two pharmaceutical agents used primarily for the management of neuropathic pain. Pregabalin is a structural analogue of gamma-aminobutyric acid (GABA) that binds to the alpha2-delta subunit of voltage-gated calcium channels in the central nervous system, thereby modulating the release of excitatory neurotransmitters such as glutamate, substance P, norepinephrine, and calcitonin gene-related peptide[6]. Tramadol is an opioid analgesic that acts as a weak agonist at the mu-opioid receptor and also inhibits reuptake of serotonin and norepinephrine. The combination has been shown to have synergistic or super-additive effects on pain relief in some studies[3][8], making it potentially more effective than either drug alone for certain types of chronic or neuropathic pain. However, combining these drugs increases the risk for central nervous system depression—including respiratory depression—and other adverse effects such as impaired coordination or coma; thus caution is advised when co-administering them[1][2][5]. Pharmacokinetic studies indicate no significant drug-drug interaction between pregabalin and tramadol when used together[4].
