Pregabalin + venlafaxine is a combination therapy being investigated by Beijing Tiantan Hospital for the treatment of moderate to severe fibromyalgia. Venlafaxine is a serotonin-norepinephrine reuptake inhibitor (SNRI) that modulates pain pathways by increasing the availability of serotonin and norepinephrine in the central nervous system. Pregabalin is a gabapentinoid that binds to the alpha-2-delta subunit of voltage-gated calcium channels, thereby reducing the release of excitatory neurotransmitters. The combination aims to provide superior pain relief compared to pregabalin monotherapy by targeting multiple pathways involved in central sensitization and neurotransmitter imbalance associated with fibromyalgia.
