Prexasertib + samotolisib is an investigational combination therapy composed of two small molecule inhibitors developed for cancer treatment. Prexasertib (LY2606368) is a checkpoint kinase 1 (CHK1) inhibitor that disrupts DNA damage response and cell cycle progression, leading to increased tumor cell death. Samotolisib (LY3023414) is a dual PI3K/mTOR inhibitor that targets the phosphoinositide 3-kinase and mammalian target of rapamycin pathways, both critical for cancer cell growth and survival. The combination has demonstrated antitumor activity in preclinical models and preliminary efficacy in heavily pretreated patients with advanced cancers[1][4]. Both agents were developed by Eli Lilly but have since been discontinued from further development[2].
