Overview

PRGN-2009 + pembrolizumab is a combination therapy under investigation for the treatment of human papillomavirus (HPV)-associated cancers, particularly recurrent or metastatic cervical cancer. PRGN-2009 is an off-the-shelf therapeutic vaccine based on a gorilla adenovirus vector that delivers HPV type 16 and 18 antigens to induce a de novo HPV-specific immune response by transgene expression in antigen-presenting cells. Pembrolizumab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, acting as an immune checkpoint inhibitor to enhance anti-tumor immunity. The combination aims to leverage both direct immunization against HPV antigens and checkpoint blockade to improve anti-tumor responses in patients with advanced or resistant disease[2][5][6].
