Overview

PRI-724 + cytarabine is a combination of **PRI-724**, a small molecule inhibitor that blocks the interaction between CREB-binding protein (CBP) and β-catenin, and **cytarabine**, a pyrimidine nucleoside analog antimetabolite chemotherapy primarily used for the treatment of leukemias. PRI-724 modulates the Wnt/β-catenin signaling pathway, potentially inhibiting self-renewal of cancer stem cells and increasing tumor sensitivity to cytotoxic drugs, while cytarabine disrupts DNA synthesis in rapidly dividing cells by inhibiting DNA polymerase, leading to cancer cell death. This combination is being investigated for its potential to overcome chemotherapy resistance in hematologic malignancies and selected solid tumors[3][5][8].
