Overview

**PRI-724 + dasatinib** is an investigational combination therapy consisting of two small molecules with distinct mechanisms of action targeting cancer and fibrosis pathways. PRI-724 (also known as foscenvivint) is a selective inhibitor of the canonical Wnt/β-catenin signaling pathway that works by specifically blocking the interaction between β-catenin and the transcriptional coactivator CREB binding protein (CBP)[1][5][7]. This modulation results in the inhibition of gene transcription essential for tumor proliferation and fibrogenesis, promoting stem cell differentiation and eliminating tumor-initiating cells. Dasatinib is an orally available multi-kinase inhibitor that blocks the activity of the BCR-ABL fusion protein and several other tyrosine kinases (SRC family, c-KIT, EPHA2, PDGFRβ), leading to decreased growth and proliferation of cancer cells[2][4][6][8]. The combination is explored for enhanced anti-tumor efficacy by acting on complementary pathways; however, specific clinical evidence for the combination "PRI-724 + dasatinib" is not currently established in published clinical trials.
