This is a combination regimen composed of PRO95780, cetuximab, and irinotecan. PRO95780 is a fully human IgG1 monoclonal antibody that acts as an agonist of death receptor 5 (DR5/TRAIL-R2/TNFRSF10B), triggering the extrinsic apoptosis pathway in tumor cells[1][5]. Cetuximab is a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling. Irinotecan is a small molecule chemotherapeutic that inhibits topoisomerase I, leading to DNA damage and cell death. Combined, these agents are designed to provide antitumor effects via apoptosis induction, EGFR inhibition, and DNA replication inhibition, respectively. Such combinations may be explored in clinical trials for cancer indications where multi-targeted therapy is desired.
