ProAgio + gemcitabine + nab-paclitaxel is an investigational combination therapy consisting of ProAgio, a novel protein-based drug (ACT50) that selectively binds to integrin αvβ3 on cancer-associated fibroblasts (CAFs) and induces their apoptosis, combined with gemcitabine, a nucleoside analog that inhibits DNA synthesis, and nab-paclitaxel (nanoparticle albumin-bound paclitaxel), a microtubule inhibitor. The combination is being studied as a first-line treatment for metastatic pancreatic ductal adenocarcinoma (PDAC), aiming to enhance antitumor activity by targeting both the tumor stroma (via ProAgio) and tumor cells (via chemotherapy). This regimen is under investigation in an open-label Phase I/Ib trial to assess safety, pharmacokinetics, and preliminary efficacy[1][3][5][6].
