Overview

Programmed Death-1 inhibitors + Programmed Death-Ligand 1 inhibitors refers to the combined use of two classes of immune checkpoint inhibitor drugs that target the PD-1 (programmed death protein 1) receptor and its ligand, PD-L1 (programmed death-ligand 1). Both are monoclonal antibodies designed to block the interaction between PD-1 on T cells and PD-L1 on tumor or other cells. This blockade prevents cancer cells from evading immune detection, thereby enhancing T-cell-mediated anti-tumor responses. These combinations are used in oncology for a variety of advanced or metastatic cancers and may be administered together or as part of multi-drug regimens. The most common agents in these classes include pembrolizumab, nivolumab, cemiplimab (PD-1 inhibitors), and atezolizumab, durvalumab, avelumab (PD-L1 inhibitors)[2][3][4][7]. Combination therapy is under investigation for synergistic effects but is not yet standard for all indications.
