Drug intelligence / Profile preview
Propafenone; flecainide; sotalol; dofetilide refers to a grouped intervention of anti-arrhythmic drugs (AADs) evaluated in the **Antiarrhythmics After Ablation of Atrial Fibrillation (5A Study)**. This prospective randomized trial, conducted by the **University of Pennsylvania**, investigated whether the empirical use of these AADs for six weeks following pulmonary vein isolation (PVI) could reduce the recurrence of atrial arrhythmias in patients with paroxysmal atrial fibrillation. The group includes Class Ic agents (propafenone and flecainide), which primarily block the **sodium channel protein type 5 subunit alpha (SCN5A)**, and Class III agents (sotalol and dofetilide), which primarily block the **potassium voltage-gated channel subfamily H member 2 (KCNH2)** to prolong the cardiac action potential. Sotalol also possesses non-selective beta-adrenergic blocking activity.
