Propofol + clonidine is a combination of two drugs used primarily in anesthesia and procedural sedation. Propofol is an intravenous anesthetic agent that acts mainly as a positive allosteric modulator of the gamma-aminobutyric acid type A (GABA-A) receptor, leading to central nervous system depression and rapid induction of unconsciousness. Clonidine is an alpha-2 adrenergic receptor agonist with sedative, analgesic, and antihypertensive properties; it reduces sympathetic outflow from the central nervous system. When used together, clonidine enhances the sedative effects of propofol, allowing for reduced propofol dosage requirements while maintaining effective anesthesia or sedation. This combination has been shown to provide faster onset of sedation, improved patient satisfaction, deeper levels of sedation, and greater hemodynamic stability compared to propofol alone; however, it may also prolong recovery times[1][2][5][6][8]. The mixture can be physically compatible for co-administration under certain conditions[4].
