Overview

This investigational combination therapy is being evaluated as a first-line treatment for advanced non-small cell lung cancer (NSCLC) that is negative for EGFR, ALK, and ROS1 driver mutations. The regimen consists of **propranolol hydrochloride**, a non-selective beta-adrenergic receptor antagonist; **sintilimab**, a recombinant humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor; and standard **platinum-based chemotherapy** (typically cisplatin or carboplatin combined with pemetrexed, paclitaxel, or gemcitabine). Propranolol is included to potentially modulate the tumor microenvironment and augment immune cell responses, thereby enhancing the efficacy of the immune checkpoint inhibitor sintilimab. The combination is currently under investigation in a Phase I clinical trial (BRIO) sponsored by the Second Xiangya Hospital of Central South University.
