Overview

This is a combination drug consisting of propylthiouracil (PTU) and thyroxine (levothyroxine, LT4). Propylthiouracil is an antithyroid agent that inhibits thyroid hormone synthesis by blocking the organification of iodine and coupling of iodotyrosines in the thyroid gland, as well as inhibiting peripheral conversion of thyroxine (T4) to triiodothyronine (T3)[4][5]. Levothyroxine is a synthetic form of the thyroid hormone T4, used to replace or supplement endogenous thyroxine in hypothyroid states. The combination has been studied for use in specific conditions such as monocarboxylate transporter 8 (MCT8) deficiency/Allan–Herndon–Dudley syndrome, where it can help normalize serum T3 and T4 levels[1]. The combined therapy requires individualized dosing adjustments.
