Overview

Proton beam irradiation + ranibizumab is a combination therapy used primarily in ophthalmology for the management and prevention of radiation-induced complications following treatment of intraocular tumors such as choroidal or uveal melanoma. Proton beam irradiation is a form of highly targeted external radiotherapy that uses protons to deliver precise doses to ocular tumors while sparing surrounding healthy tissue. Ranibizumab is a recombinant humanized monoclonal antibody fragment (Fab) that binds and inhibits vascular endothelial growth factor A (VEGF-A), thereby preventing abnormal blood vessel growth and leakage in the retina. The combination has been studied for its ability to reduce the incidence and severity of radiation retinopathy or maculopathy after ocular tumor radiotherapy by inhibiting VEGF-mediated neovascularization and vascular permeability[1][5]. Clinical studies have shown that prophylactic intravitreal administration of ranibizumab during or after proton beam therapy can help preserve visual acuity and reduce retinal complications compared to historical controls[1][5].
