Overview

PRS-343 + atezolizumab is an investigational combination therapy being evaluated for the treatment of HER2-positive advanced or metastatic solid tumors. PRS-343 is a bispecific fusion protein that targets both Tumor necrosis factor receptor superfamily member 9 (4‑1BB), a co-stimulatory immune receptor, and Receptor tyrosine-protein kinase erbB‑2 (HER2), a tumor-associated antigen. It comprises a 4-1BB-targeting Anticalin protein fused to a HER2-targeting antibody, designed to activate tumor-specific T lymphocytes selectively within the tumor microenvironment. Atezolizumab (marketed as Tecentriq) is an approved monoclonal antibody that inhibits Programmed death-ligand 1 (PD-L1), thereby enhancing anti-tumor immune responses. The combination aims to synergistically enhance T cell activation and anti-tumor immunity by simultaneously engaging both pathways. Clinical studies have shown evidence of durable clinical benefit in heavily pre-treated patients with various HER2-positive cancers, including complete and partial responses[2][3][5][6].
