Overview

**PRT1419 + azacitidine** is an investigational combination therapy currently in early-phase clinical studies for the treatment of relapsed or refractory myeloid malignancies. **PRT1419** is a potent small molecule inhibitor targeting myeloid cell leukemia-1 (Mcl-1), an anti-apoptotic BCL-2 family protein implicated in cancer cell survival. By inhibiting Mcl-1, PRT1419 promotes apoptosis in malignant cells. **Azacitidine** is a hypomethylating agent and cytidine nucleoside analog that is commonly used in the treatment of myelodysplastic syndromes and acute myeloid leukemia; it works primarily by incorporating into DNA and RNA to cause hypomethylation and direct cytotoxicity to abnormal hematopoietic cells. This combination aims to utilize both direct pro-apoptotic (via Mcl-1 inhibition) and epigenetic (via DNA hypomethylation) mechanisms to target resistant disease forms[1][3][5][7][8][9].
