Overview

PRT2527 + venetoclax is an investigational combination therapy being studied in relapsed or refractory hematologic malignancies, particularly myeloid malignancies. PRT2527 is a potent and highly selective cyclin-dependent kinase 9 (CDK9) inhibitor, which suppresses transcription of key anti-apoptotic proteins such as MCL1 and MYC, leading to apoptosis in cancer cells. Venetoclax is a small molecule inhibitor of B-cell lymphoma 2 (BCL2), which induces apoptosis by directly inhibiting the anti-apoptotic BCL2 protein. The rationale for this combination is that simultaneous inhibition of CDK9 (lowering MCL1 and MYC) and BCL2 induces synergistic cell death and can overcome resistance to BCL2 inhibition frequently observed in hematologic cancers[1][3][2]. This combination is being studied in phase 1 clinical trials in several hematologic malignancies, including acute myeloid leukemia, myelodysplastic syndrome, and others[1][2][3].
