Overview

Prucalopride + polyethylene glycol is a combination therapy consisting of two agents used for the management of chronic constipation, particularly in patients who have not responded adequately to standard laxative therapy. Prucalopride is a highly selective serotonin type 4 (5-HT4) receptor agonist that stimulates colonic motility by enhancing acetylcholine release from enteric neurons, thereby increasing the frequency and amplitude of colonic contractions and promoting bowel movements. Polyethylene glycol (PEG) is an osmotic laxative that increases water content in the stool by retaining fluid in the colon, resulting in softer stools and increased stool frequency. The combination aims to leverage both mechanisms—direct stimulation of gut motility (prucalopride) and improved stool consistency/volume (PEG)—to improve outcomes for patients with refractory chronic constipation[1][2][5][6].
