Overview

PSCA-CAR T cells (CD28) are an adoptive cellular immunotherapy consisting of T cells (typically autologous) engineered to express a chimeric antigen receptor (CAR) targeting the prostate stem-cell antigen (PSCA). This specific CAR construct utilizes a CD28 co-stimulatory domain in conjunction with a CD3-zeta signaling domain. PSCA is a cell-surface glycoprotein that is highly overexpressed in prostate cancer, particularly in bone metastatic and castration-resistant disease, as well as in some pancreatic and bladder cancers. The inclusion of the CD28 domain is designed to provide robust and rapid T-cell activation and effector function upon binding to the target antigen. Research conducted at City of Hope has compared these constructs to 4-1BB-based CARs, noting that while CD28-based CARs are highly potent, they may exhibit different persistence and exhaustion profiles in the immunosuppressive microenvironment of solid tumors.
