Overview

CRx-150 is a fixed-dose combination drug candidate developed by CombinatoRx, consisting of the nasal decongestant pseudoephedrine and the nonsteroidal anti-inflammatory drug (NSAID) naproxen. It was designed to provide symptomatic relief for conditions such as the common cold, influenza, and allergic rhinitis by simultaneously addressing nasal congestion and associated pain or inflammation. Pseudoephedrine acts as a sympathomimetic agent, primarily serving as an agonist at alpha-adrenergic receptors to induce vasoconstriction in the respiratory mucosa, thereby reducing swelling and congestion. Naproxen provides analgesic and anti-inflammatory effects by inhibiting cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) enzymes, which reduces the synthesis of pro-inflammatory prostaglandins. The product was identified using CombinatoRx's proprietary high-throughput screening approach to find synergistic pairings of existing medications, but development was ultimately discontinued during the mid-2000s.
