Overview

**Psilocybin** is a naturally occurring, orally administered serotonergic psychedelic small molecule and prodrug of psilocin. Psilocin primarily stimulates the serotonin 2A receptor, producing acute psychedelic effects and potentially facilitating psychotherapy-associated changes in cognition and emotional processing. Unity Health Toronto is studying a single 25 mg dose of psilocybin as an investigational adjunct to massed cognitive processing therapy in an open-label Phase 2 feasibility trial in 15 adults with chronic posttraumatic stress disorder. The study evaluates feasibility, tolerability, safety, and preliminary effects on PTSD severity; psilocybin is not a marketed PTSD treatment in this trial context.
