Overview

This therapeutic intervention consists of a combination of synthetic psilocybin and transcutaneous auricular vagus nerve stimulation (taVNS) for the treatment of major depressive disorder (MDD). Psilocybin is a serotonergic psychedelic that acts as a prodrug for psilocin, which primarily targets the 5-HT2A receptor to facilitate rapid neuroplasticity and profound psychological shifts. The taVNS component is a non-invasive medical device that delivers electrical stimulation to the auricular branch of the vagus nerve, aiming to modulate autonomic tone and enhance the therapeutic window provided by psilocybin. Developed by Dr. Charles Raison and investigated in the OPTIMIZE Phase 2 clinical trial, this approach explores whether neuromodulation can sustain or amplify the antidepressant effects of a single 25 mg psychedelic dose.
