Psilocybin + trazodone is an experimental combination therapy being investigated by Centre Hospitalier St Anne for the treatment of treatment-resistant depression (TRD). Psilocybin is a prodrug of psilocin, a potent agonist at the serotonin 5-HT2A receptor, known for its rapid antidepressant effects and neuroplasticity-inducing properties. Trazodone is an antidepressant that acts as a 5-HT2A receptor antagonist. This combination is being evaluated in the Phase 2 PSILOTRAZ trial to determine if the therapeutic antidepressant benefits of psilocybin can be decoupled from its acute subjective psychedelic effects by blocking the 5-HT2A receptor during administration. The psilocybin component used in this specific study is PEX010, a botanical extract developed by Filament Health.
