PSMA+GD2 CAR-T is a chimeric antigen receptor T cell (CAR-T) therapy designed to target both Prostate-Specific Membrane Antigen (PSMA) and disialoganglioside GD2. This dual-targeting approach aims to enhance antitumor efficacy by recognizing and attacking tumor cells expressing either or both of these surface antigens. PSMA is highly expressed in prostate cancer and some other solid tumors, while GD2 is overexpressed in neuroblastoma, melanoma, retinoblastoma, Ewing sarcoma, and certain gliomas. The therapy involves engineering patient-derived T cells to express synthetic receptors that bind specifically to these tumor-associated antigens, leading to targeted cytotoxicity against cancer cells. The mechanism of action includes direct tumor cell lysis via T-cell activation upon antigen engagement as well as the induction of proinflammatory cytokines within the tumor microenvironment.
