Overview

PSMA | PLP1 Tmod CAR-T is an autologous T-cell therapy developed by A2 Biotherapeutics for the treatment of prostate cancer. It utilizes the company's proprietary Tmod (T-cell module) platform, which incorporates a synthetic NOT gate logic system to enhance safety and specificity. The CAR-T cells are engineered with an activator receptor targeting Prostate-Specific Membrane Antigen (PSMA) and an inhibitory receptor targeting Proteolipid Protein 1 (PLP1). While PSMA is highly expressed in prostate cancer, it is also present in the central nervous system (CNS), specifically in human oligodendrocytes and Schwann cells. PLP1, an integral membrane protein of the myelin sheath, is co-expressed with PSMA in these CNS tissues. The Tmod system allows the CAR-T cells to potently kill PSMA-positive tumor cells while sparing healthy CNS tissues that co-express PLP1, thereby addressing the potential risk of on-target, off-tumor neurotoxicity.
