Pt(0506)101 is a novel platinum-based antineoplastic small molecule developed by Beijing Shuobai Pharmaceutical Technology. As a platinum complex, it is designed to exert cytotoxic effects by covalently binding to DNA, primarily forming intra-strand and inter-strand cross-links. These structural modifications distort the DNA helix, effectively blocking DNA replication and RNA transcription, which triggers cell cycle arrest and programmed cell death (apoptosis). Pt(0506)101 is currently in Phase Ib/IIa clinical development in China, where it is being evaluated as a first-line treatment for patients with unresectable locally advanced, recurrent, or metastatic HER2-negative, PD-L1-positive gastric or gastroesophageal junction (GEJ) adenocarcinoma in combination with capecitabine and the PD-1 inhibitor sintilimab.
