PT-112 + gemcitabine is an investigational combination therapy comprising PT-112, a novel pyrophosphate-platinum conjugate, and gemcitabine, a nucleoside metabolic inhibitor. PT-112 is a small molecule chemotherapeutic agent designed to induce immunogenic cell death (ICD) and possesses osteotropic (bone-targeting) properties due to its pyrophosphate moieties. Gemcitabine acts by inhibiting DNA synthesis and ribonucleotide reductase. The combination is primarily being evaluated for the treatment of advanced solid tumors, with a specific focus on biliary tract cancer. It is currently in Phase I/II clinical development, particularly in trials conducted in China (e.g., NCT05357196).
