Drug intelligence / Profile preview
**PTEN mRNA@HA-LNP** is an experimental, topically administered mRNA nanomedicine developed in an academic preclinical study for localized melanoma immunotherapy. It encapsulates chemically modified messenger RNA encoding the tumor-suppressor phosphatase and tensin homolog protein in a hyaluronate-conjugated lipid nanoparticle containing hyaluronic acid-dimyristoyl glycerol. The hyaluronate surface component replaces PEG-like stabilization functionality, promotes skin and tumor penetration, and supports CD44-mediated uptake by CD44-expressing tumor cells. Following intracellular delivery, the mRNA transiently restores PTEN protein expression, suppressing PI3K-AKT signaling, reducing melanoma-cell viability, inducing immunogenic cell death, and enhancing antitumor immune activation in melanoma models. The reported evidence is preclinical, including in vitro experiments and topical treatment of melanoma-bearing mice.
