PTEN mRNA@PEG-LNP is an experimental mRNA-based therapeutic formulation designed to restore the expression of the Phosphatase and tensin homolog (PTEN) protein. It consists of messenger RNA (mRNA) encoding the PTEN tumor suppressor, encapsulated within lipid nanoparticles (LNPs) that are functionalized with polyethylene glycol (PEG) for structural stability and biocompatibility. PTEN is a critical negative regulator of the PI3K/Akt/mTOR signaling pathway; its loss or inactivation is a common driver in various cancers, including melanoma, leading to uncontrolled cellular proliferation and resistance to immune checkpoint inhibitors. By delivering exogenous PTEN mRNA, this formulation aims to reactivate antitumor immunity, induce immunogenic cell death, and inhibit tumor progression. In research contexts, it often serves as a benchmark for comparison with next-generation delivery systems, such as hyaluronated lipid nanoparticles (HA-LNPs), which are designed for enhanced skin penetration or targeted delivery.
