This is a combination of two peptide hormones, parathyroid hormone fragment 1–34 [PTH(1–34)] and parathyroid hormone-related protein fragment 1–36 [PTHrP(1–36)]. Both peptides act as bone anabolic agents by stimulating bone formation. They exert their effects primarily through activation of the same G protein-coupled receptor, the parathyroid hormone/parathyroid hormone-related peptide receptor (PTHR). While both ligands bind to and activate this receptor, they differ in their binding kinetics and signaling duration. PTH(1–34) binds more strongly to the G protein-uncoupled conformation of the receptor, resulting in longer-lasting signaling compared to PTHrP(1–36), which dissociates more rapidly. This combination has been studied for its potential to increase bone mineral density and treat conditions such as osteoporosis[3][4][1].
