PTK7 + CPT113 is an investigational antibody-drug conjugate combination targeting Protein tyrosine kinase 7 (PTK7) using the CPT113 linker-payload platform delivering a topoisomerase I inhibitor payload; it is being advanced for high PTK7-expressing solid tumors such as non-small cell lung cancer and ovarian cancer. According to ADC Review and company disclosures, the CPT113 platform features a highly stable cleavable linker designed to release a topoisomerase I inhibitor payload, positioning PTK7-CPT113 as part of a next-generation ADC portfolio in-licensed by Aadi Bioscience from Hangzhou DAC for development in PTK7-high cancers.[7][1][10] PTK7 is a catalytically inactive receptor in the non-canonical Wnt pathway implicated in tumor biology and has been clinically explored as an ADC target with earlier auristatin-based PTK7 ADCs (e.g., cofetuzumab pelidotin) showing activity in ovarian cancer, NSCLC, and TNBC, supporting the rationale for a PTK7-directed topoisomerase I ADC approach.[2][4][5]
