Overview

PTPRC-CYRIB fusion gene (lentiviral overexpression) is an experimental genetic construct used to investigate the mechanisms of drug resistance in T-cell acute lymphoblastic leukemia (T-ALL). The PTPRC-CYRIB fusion gene was identified via next-generation sequencing in a patient with primary drug-resistant T-ALL and subsequently validated through molecular cloning. By overexpressing this fusion gene in T-ALL cell lines using a lentiviral vector, researchers demonstrated that it significantly increases cell proliferation rates and enhances clonogenic potential. Furthermore, overexpression of PTPRC-CYRIB has been shown to confer resistance to standard chemotherapeutic agents, including vincristine, idarubicin, cyclophosphamide, and prednisone, suggesting its role as a critical driver of chemoresistance in this leukemia subtype.
