Overview

PU-H71 + nab-paclitaxel is a combination therapy being investigated for cancer treatment, particularly in HER2-negative metastatic breast cancer. PU-H71 is an epichaperome inhibitor that binds to the ATP-binding site of HSP90, disrupting its chaperone activity and leading to degradation of client proteins involved in oncogenic signaling pathways. Nab-paclitaxel (Abraxane) is an albumin-bound formulation of paclitaxel that promotes assembly and stabilization of microtubules, inhibiting normal dynamic reorganization of the microtubule network essential for cell division. In clinical trials, this combination has shown evidence of clinical activity with more durable disease control observed in patients with high baseline epichaperome expression.
