Overview

Pucotenlimab (AK106) is a humanized IgG1 monoclonal antibody targeting CD47, a cell surface protein often overexpressed on cancer cells. CD47 acts as a "don't eat me" signal by interacting with SIRPα on macrophages, thereby inhibiting phagocytosis. By blocking the CD47-SIRPα interaction, pucotenlimab enhances the phagocytic clearance of tumor cells by macrophages and may also promote an adaptive immune response. Developed by Akeso Biopharma, it is engineered to have low affinity for red blood cells to minimize hemagglutination and anemia, which are common side effects of CD47 inhibitors. It is currently being investigated for the treatment of various solid tumors and hematological malignancies, both as a monotherapy and in combination with other agents.
