Pucotenlimab + lenvatinib is an investigational combination therapy consisting of pucotenlimab, a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), and lenvatinib, a small molecule multi-kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR1–3), fibroblast growth factor receptors (FGFR1–4), platelet-derived growth factor receptor alpha (PDGFRα), RET proto-oncogene, and KIT proto-oncogene. Pucotenlimab acts as an immune checkpoint inhibitor to enhance anti-tumor immune responses by blocking the PD-1/PD-L1 pathway. Lenvatinib inhibits angiogenesis and tumor proliferation through its action on multiple tyrosine kinases involved in tumor vascularization and growth. The combination is being studied primarily for advanced hepatocellular carcinoma (aHCC) and unresectable intrahepatic cholangiocarcinoma as conversion or neoadjuvant therapy[2][3][5][7].
