Overview

This combination therapy integrates the humanized anti-PD-1 monoclonal antibody pucotenlimab with the multikinase inhibitor lenvatinib and the alkylating chemotherapy agent temozolomide. It is specifically being evaluated in a Phase II clinical trial (PLT-NAT-HNMM-II) led by Shanghai Ninth People's Hospital for the neoadjuvant and adjuvant treatment of resectable head and neck mucosal melanoma. Pucotenlimab works by inhibiting the PD-1/PD-L1 checkpoint, thereby reactivating the anti-tumor immune response. Lenvatinib targets multiple receptor tyrosine kinases, including VEGFR1-3 and FGFR1-4, to inhibit tumor angiogenesis and lymphangiogenesis. Temozolomide provides cytotoxic activity by alkylating DNA, leading to cell death. The triplet regimen aims to improve the prognosis of mucosal melanoma, which is typically resistant to conventional therapies.
